LightCycler® probe 5'-LC dye and 3'-phosphate

Hybridization probes consist of a 3’- fluorescein and a 5'-LightCycler® Red labeled pair of oligonucleotide probes. The fluorescein transmits its energy onto the acceptor dye. The generated FRET signal is proportional to the amount of the bonded single stranded target.

The 5’ labeled probe is modified at the 3’ end to prevent its extension by the Taq polymerase during the PCR process.

FRET Hybridization probes work with LightCycler® and RotorGene instruments. Appropriate acceptor dyes are:

• LightCycler® Red 610, ROX, Texas Red (LC 2.0, LC480 und RotorGene)

• LightCycler® Red 640 (alle Geräte)

• LightCycler® Red 670, Cy5 (LC 2.0, LC480 und RotorGene)

• LightCycler® Red 705, Cy5.5 (LC 1.x und 2.0)

FRET Hybridization probes can be used for quantification and genotyping. Genotyping results from analyzing the melting curve of the PCR product. This technique of genotyping is more precise than the analysis through TaqMan® probes, because unknown variants can be detected. TaqMan® probes also emit fluorescence in the absence of a variant and can, therefore, only be used for biallelic systems.
